I'm looking into iSCSI for the first time and the first rule seems to be not to use the same target on multiple computers. Does this rule also apply if you map the drive on another computer instead of using iSCSI? I want to use a local drive as an iSCSI drive on a VM running on my server but also have network access to it from my workstation.

You can use the same iSCSI Target on multiple computers, but you need to ensure data consistency and avoid conflicts. If multiple initiators attempt to write to the same storage area simultaneously, it may lead to data corruption or other unpredictable problems.
