Hello, i find on my u4-500 free pcie x4 slot, he is gen4 ? possible add on this slot another nvme or 25gbps NIC ?

If it's not behind a 600 series PCH on the motherboard, then yes, it's Gen4×4, right from the CPU itself.
Technically, you could add anything into the slot (with an extension since the 1U height would limit vertical install), however TOS might not recognize it if drivers aren't available/installed for it; if you're running an alternate OS & drivers exist or can be injected/installed, then you can use whatever you've installed.

The U4-500 has a reserved standard PCIe slot position inside the chassis. The physical form factor of the slot is PCIe x4, while the actual signal runs at PCIe 3.0 x2. Due to the structural constraints of the U4-500 enclosure, no PCIe expansion devices can be installed in this slot.
