Backup to External USB Drive... Hyper Backup? USB Copy? NTFS? EXT4? exFAT?

This is a question for a new small business file server that will be keeping mostly our database of picture files.
Our current Windows server is running our domain controller, DNS, and file server, but we are wanting to separate our file server.

We purchased an external hard drive (16TB) that has its own power source (to prevent brownouts) to serve as a backup to the NAS.

For our purposes and our files, what is the best solution for backing up?
From my research, it looks like Hyper Backup would be the correct solution, but are there different ones that would possibly provide more flexibility with how the backup is used in case it is needed?

What file system would be recommended for the backup drive? I was thinking NTFS so we can plug the backup drive into a Windows workstation if need be, but is a Hyper Backup backup even readable?

You can download USB Copy in the application center to back up data to an external storage device.
You can choose the file system you want based on the advantages of different file systems.
